Tulathromycin is a third-generation macrolide antibiotic. It has two isomers: a 13-membered azalide ring and a 15-membered azalide ring. They form a stable mixture in a 1:9 ratio and are interconvertible. Tulathromycin possesses three basic amino groups, which distinguish it from azalides, ketolides, and other macrolides. Because of these three basic amino groups, the molecule carries a high positive charge and exhibits a strong electronegativity in solution. This electrostatic charge enhances the drug's ability to penetrate the outer membrane of lipopolysaccharide (LPS), favoring it for penetration of the outer membrane of Gram-negative bacteria, resulting in a strong antibacterial effect.
